Generation of Monoclonal Antibodies Reactive to Human Interleukin 2(IL2)

Abstract

|
Monoclonal antibodiesa(mAb) were generated from clones that hybrid of P3×63 Ag 8.653 with spleen cells of BALB/c mouse immunized with IL 2(human recombinant IL 2). Among hybridoma, mAb producing nie clones were selected by ELISA screening
method.
These
mAb detected by ELISA were found to react specifically with IL 2 as demonstrated by antibod-dependent neutralization of IL 2 activity. Class and isotype of mAb were identified also by ELISA of peroxidaseconjugated anti-mouse Ig. These results
showed
IgM(one clone), IgG1(4 clones), IgG2a(3 clones) and IgG2b(one clone). MAbs produced in vivo were purified by affinity chromatography and anion exchange chromatography in FPLC, and identified by SDS-PAGE and Ouchterlong gel diffusion methods.
